Confirmed in an interview by Cinema Blend that WWE Star John Cena to star as Duke Nukem himself.
As the film doesn’t even have a script for this upcoming film, it’s safe to say that Cena will be the alien fighter. As said in the interview by producer Andrew Form and Brad Fuller with Cinema Blend:

Yeah, that’s what we’re working with now. He is. Yes. We don’t have a script yet, so that is confirmed at this point but if he reads the script and he doesn’t like the script I’m sure there’s ways that he could pull out, but right now he’s our guy.

Until the script is made and nothing in between, Cena will be the star of the film. Along with the interview, it was said by the producers that they were looking to follow the same tone as the recent Deadpool movie was. As many already know, there’s low hope for videogame movies being successful and trying to make the film less serious especially for Duke Nukem could actually be a good turn of events for the genre’s current status.
